In order to get it working in DOS, make sure the vacsid.ini settings are correct. By default the sound configuration is set to AMD Interwave with a frequency of 64000.
For Sound Blaster, just set "device" to 6 with a frequency to 44100.

I couldn't get VacSID working in DOSBox. I was only able to run it with VirtualBox with an image of MSDOS 6.22.
